The Indiana University O'Brien National Resource Center (NRC) is part of the George M. O'Brien Kidney Resource Alliance (OKRA). From its inception as an NIDDK-funded P50 Center in 2002, the mission of the IU O’Brien NRC has been to provide renal researchers with effective access to advanced forms of microscopy. Over the past 20+ years, the IU O'Brien NRC has pioneered the development and dissemination of novel methods of multiphoton intravital microscopy, quantitative tissue cytometry, fixed tissue imaging, and digital image analysis that have been applied in collaborative studies of renal physiology and pathophysiology with investigators from around the world. Studies conducted with the IU O’Brien NRC have generated data used in over 200 publications and numerous grant applications that have helped launch the careers of a new generation of investigators. The goals of the IU O’Brien Center are to: 1. Provide quantitative intravital microcopy of mice and rats as a service to renal researchers. 2. Provide quantitative, large-scale microscopy of the kidney as a service to renal researchers through a combination of 3D confocal tissue cytometry, CODEX multiplexed cytometry and spatial transcriptomics, thus delivering uniquely comprehensive molecular characterizations. 3. Develop methods of tissue clearing, light-sheet microscopy and image analysis to expand the scale of quantitative microscopy of renal tissues. These services, designed to extend the scope and power of renal research studies are supported through an Opportunity Pool Program supported by the OKRA National Coordinating Center and a Pilot and Feasibility Program supported by Indiana University. The center also supports education through a unique student enrichment program and internationally-recognized advanced microscopy workshops.
