Description

The mission of the Mayo Clinic Robert M. and Billie Kelley Pirnie Polycystic Kidney Disease (PKD) Center is to translate basic scientific findings into better treatments for people with PKD. The Center spans basic research to the delivery of comprehensive care. Mayo Clinic, Rochester is the recipient of 2 Clinical Centers of Excellence designations by the PKD Foundation for adult and pediatric clinics. Clinical services include innovative treatments and clinical trials. We collaborate and share materials, resources, and expertise with both academic and industry partners to advance PKD research. Focused areas of expertise include Genetics, Epigenetics, Metabolomics, Imaging, and Model Systems. Research projects include investigation of disease origins, therapeutic pathways, biomarker identification. The Center also oversees the PKD Foundation ADPKD Mutation Database, which evaluates PKD1 and PKD2 gene variants for pathogenicity. Imaging services include advanced preclinical and clinical imaging protocols using 3D ultrasound, CT, and quantitative MRI. Cutting-edge image processing techniques include radiomics, machine learning, and deep learning. These techniques enable automated and precise analysis of organ volume, cyst number, and size, delivering patient-specific, image-based metrics. PKD models maintained and shared by the Center include C. elegans, zebrafish, and rodents. Standardized protocols for zebrafish and rodent preclinical testing are utilized to evaluate the efficacy of novel therapies and zebrafish allow for higher-throughput drug and genetic screening. Novel therapeutic approaches are examined, including base editing-mediated gene therapy and RNA-based treatments for PKD. The Center invests heavily in training the next generation of scientists, offering opportunities such as Summer Undergraduate Research Fellowships (supported by an R25 grant) and Postdoctoral Fellowships (supported by an U2C grant) to nurture emerging talent from undergraduate students through junior faculty. Finally, the Center is a component of the PKD RRC Cell Models Core contributing to the collection and distribution of PKD patient samples.
